СРАВНИТЕЛЬНЫЙ АНАЛИЗ РАЗЛИЧНЫХ МОДЕЛЕЙ АМИЛОИДОЗА

Аннотация
Рассмотрены естественные и экспериментальные модели амилоидоза в контексте существующих теорий и известных механизмов амилоидогенеза. Имеющиеся клинические и экспериментальные наблюдения свидетельствуют, что мнение о фатальной неизлечимости амилоидоза неверно. Показано, что существует значительное количество экспериментальных достаточно легко воспроизводимых моделей амилоидоза, которые могут быть использованы для отработки методов лечения этой патологии. Предложена классификация моделей амилоидоза: естественные (животные модели с генуинным амилоидозом), клеточные клоны, артифициальные (инфекционные, белковые, прочие). На основании анализа существующих моделей амилоидоза сделан вывод о том, что ни одна из принятых в научном сообществе теорий образования амилоида не объединяет и не объясняет все известные факты о механизмахамилоидогенеза. Предположено, что существует группа белков, структура β-листа которых потенциально способна к образованию амилоиднойконформации, и что β-листы таких белков имеют близкий аминокислотный состав. Условие образования амилоидной конформации — это попадание такого белка в достаточном количестве в несвойственное ему место, где ионная сила тканевой жидкости такова, что способствует образованию амилоидной конформации. По-видимому, неблагоприятная величина ионной силы среды окружения амилоидного белка обусловлена влиянием полисахаридов, тубулиновых белков и ионизированного кремния.
